This grant is focused on supporting research that provides a deeper mechanistic insight and establishes causal relationships between TDP-43 pathology and its phenotypic outcomes. It emphasizes the need for studies on the mechanistic interactions between TDP-43 and other co-pathologies like TMEM106B. Moreover, the grant encourages comparisons across various domains (mechanistic, molecular, structural, cellular, genetic, -omic, anatomical, neuropathological, etc.) among different TDP-43 proteinopathies, including but not limited to Limbic-predominant Age-related TDP-43 Encephalopathy (LATE), Frontotemporal Dementia (FTD), Amyotrophic Lateral Sclerosis (ALS), and their overlaps with or without Alzheimer's disease neuropathologic change (AD-NC).
